Removal of Zona Pellucida

INTRODUCTION

The zona pellucida must be removed from mouse embryos before aggregation for chimera production can be performed. This protocol describes a simple method for removal of the zona pellucida of preimplantation mouse embryos using an acidified Tyrode’s solution.


MATERIALS

Reagents

recipe Acidic Tyrode’s solution at room temperature

recipe KSOM-AA medium or other embryo culture medium
