INTRODUCTION
The zona pellucida must be removed from mouse embryos before aggregation for chimera production can be performed. This protocol describes a simple method for removal of the zona pellucida of preimplantation mouse embryos using an acidified Tyrodes solution.
MATERIALS
Reagents
Acidic Tyrodes solution at room temperature
It is also available from Sigma or Specialty Media
Embryos, preimplantation-stage: see Step 1 for details
KSOM-AA medium or other embryo culture medium
M2 or other HEPES-buffered
