INTRODUCTION

The final stage of cDNA library construction involves optimizing ligation of the size-fractionated cDNA to bacteriophage {lambda} arms, followed by packaging and plating of the recombinant bacteriophages.

MATERIALS

E. coli strain, freshly prepared overnight cultures

Use strains C600 (BNN93) for growth and BNN102 (C600hflA) for screening of cDNA libraries constructed in bacteriophage {lambda}gt10. Use strain Y1090hsdR for growth and screening of cDNA libraries constructed in bacteriophage {lambda}gt11. Use strain BB4 for growth and screening of cDNA libraries constructed in {lambda}ZAP and {lambda}ZAPII or strain XL1-Blue . . . [Full Text of this Article]
