INTRODUCTION
Radiolabeled oligonucleotides can be separated from unincorporated radiolabel by chromatography on silica gel resins. The protocol is suitable only for purifying oligonucleotides carrying 5'-phosphate groups. The method described in Purification of Synthetic Oligonucleotides by Polyacrylamide Gel Electrophoresisshould be used to purify oligonucleotides with free 5'-hydroxyl groups.
MATERIALS
Acetonitrile (5%, 30%, and 100%)
