INTRODUCTION
The introduction of commercial instruments, such as BIAcore, that are capable of measuring surface plasmon resonance (SPR), has simplified the study of macromolecular interactions by providing a format that may be used to measure molecular interactions in real time with small analytical amounts of material. This protocol describes the sequence of steps for analyzing a typical antibody-antigen system in a BIAcore experiment. The antibody (antithyroid stimulating hormone [anti-TSH]) and its ligand (TSH) are available commercially and can be used to optimize the parameters of the system.
