INTRODUCTION

This protocol describes the preparation of genomic DNA from a 5-ml liquid culture of yeast cells.


MATERIALS

Reagents

recipe caution Ammonium acetate, 5M

Appropriate yeast cultures

caution Chloroform:isoamyl alcohol(24:1, v:v)

recipe EDTA-Sark

Ethanol, 95%, cold

Ethanol, 70%

Growth medium, rich or selective (see Step 1)

recipe Yeast lysis buffer II

recipe NaCl, 5 M

recipe caution Phenol , equilibrated with H2O
