INTRODUCTION

This protocol and Hybridization of Bacteriophage DNA on Filters, which are based on a method described by Benton and Davis (1977), are used to identify and isolate recombinants containing DNA sequences of interest.


MATERIALS

recipe 2x SSPE

Depending on the number of filters to be processed, 1 liter or more may be required.

E. coli plating bacteria

Prepared as described in Plating Bacteriophage {lambda}.

Bacteriophage {lambda} library

Prepared as described in Ligation of Bacteriophage {lambda} Arms to Fragments of Foreign Genomic DNAor purchase from a commercial source.

recipe Denaturation solution
