Preparation of DNA from Cells in 24-Well Tissue Culture Plates

Andras Nagy, Marina Gertsenstein, Kristina Vintersten and Richard Behringer

This protocol was adapted from "Introduction of Foreign DNA into Embryonic Stem Cells," Chapter 10, in Manipulating the Mouse Embryo, 3rd edition, by Andras Nagy, Marina Gertsenstein, Kristina Vintersten, and Richard Behringer. Cold Spring Harbor Laboratory Press, Cold Spring Harbor, NY, USA, 2003.

INTRODUCTION

The original method for picking, replicating, and freezing embryonic stem (ES) colonies was based on 24-well plates. It is now used only when a relatively small number of colonies need to be processed. This protocol describes a method of DNA preparation from ES cells in 24-well plates. It is modified, with permission, from Wurst and Joyner (1993).
