INTRODUCTION
This protocol (modified from Hoffman and Winston 1987) describes the purification of genomic DNA from yeast cells. It is an adaptation of Isolation of Plasmid DNA from Yeast Cells: A Ten-Minute Preparation.
MATERIALS
Reagents
Ammonium acetate, 4 M
Appropriate yeast culture
Ethanol
Phenol:chloroform:isoamyl alcohol (25:24:1)
RNase A in potassium acetate
