Isolation of Yeast Genomic DNA for Southern Blot Analysis

INTRODUCTION

This protocol (modified from Hoffman and Winston 1987) describes the purification of genomic DNA from yeast cells. It is an adaptation of Isolation of Plasmid DNA from Yeast Cells: A Ten-Minute Preparation.


MATERIALS

Reagents

recipe Ammonium acetate, 4 M

Appropriate yeast culture

Ethanol

caution Phenol:chloroform:isoamyl alcohol (25:24:1)
