INTRODUCTION
There are numerous methods available for the isolation of yeast DNA. This protocol describes the preparation of DNA from a 5-ml liquid culture of yeast.
MATERIALS
Reagents
Appropriate yeast culture
Isopropanol
Potassium acetate, 5 M
RNase A in potassium acetate solution (1 mg/ml) (optional, see Step 13)
SDS, 10%
Sodium acetate, 3 M
Sorbitol, 1 M containing 0.1 M Na2 EDTA (pH 7.5)
