INTRODUCTION
There are numerous methods for the isolation of DNA from yeast cells. This protocol describes the preparation of DNA from a 40-ml liquid culture.
MATERIALS
Reagents
Appropriate yeast culture
Ethanol, 95%
Isopropanol
Potassium acetate, 5 M
RNase A in potassium acetate solution (1 mg/ml)
SDS, 10%
Sorbitol, 0.9 M, containing 0.1 M Na2 EDTA (pH 7.5)
TE (pH 7.4)
Tris-Cl, 50 mM (pH 7.4) containing 20 mM Na2 EDTA
